Our "One-Stop Metabolite Shop" concept comprises a comprehensive suite of technologies to enable synthesis of a wide range of metabolite types arising from a variety of pathways:
- CYP and non-CYP Phase I mechanisms, including AO and FMO
- Phase II conjugates such as glucuronides, glucosides and sulphates
- Mixed and multi-step pathways
We use a variety of methods to maximise the chances of success, including chemical synthesis, microbial biotransformation, mammalian liver fractions (S9/microsomes), recombinant enzymes, as well as the direct purification of metabolites from biological matrices.
Clients regularly ask Hypha to produce metabolites for unambiguous identification, for use as quantitation standards or larger amounts for pharmacology and toxicology studies to satisfy regulatory guidelines.
Hypha also offers kits for synthesis of metabolites in the client's own labs. Hypha's PolyCYPs+ screening and scale-up kits are available for making phase I metabolites of drugs and agrochemicals derived from cytochrome P450 enzymes, aldehyde oxidase and flavin-containing monooxygenase 3. The kits have also been successfully used for making GSH conjugates by incubating reactions in the presence of glutathione. Reactions can be scaled up to obtain material for structure elucidation by NMR spectroscopy.
All reagents are provided as lyophilised powders in sealed vials together with a 24-well reaction plate and air-permeable seal. Everything needed to perform the reactions is included in the kit.
